The prevalence of small ruminant lentivirus (SRL) infection was evaluated in goats and sheep in two counties of Pernambuco State, Brasil. Seriological examinations were performed from a total of 672 goats and 325 sheep, one year of age and older, at two abattoirs. Sera were analyzed by agar gel immunodiffusion using Maedi-Visna K-1514 antigens. There were 42 reactive samples (95% confidence interval 3.6% to 4.9%) in both slaughter houses. In São Lourenço da Mata county, 3.2% and 4.0% of goat and sheep sera were responsive, whereas in Paulista county, 5.1% of goat samples and 8.2% of sheep were reactive. Thus, the prevalence of small ruminant lentivirus was low in goats and sheep sampled from the region. |

O objetivo deste trabalho foi analisar amostras de soro e de células sangüíneas de caprinos para detecção de anticorpos e DNA proviral do vírus da artrite-encefalite caprina (CAEV), respectivamente. Utilizou-se a técnica de imunodifusão em ágar (AGID) e a reação em cadeia da polimerase (PCR) com "primers" degenerados. Foram analisadas amostras de diferentes procedências: 39 de Mato Grosso do Sul (MS), 19 de São Paulo (SP) e 22 do Ceará (CE), dessas últimas, 12 oriundas de animais importados do Canadá. Os resultados de AGID e PCR foram discordantes, pois o primeiro permitiu a detecção de 25 animais soropositivos, enquanto a PCR detectou DNA proviral de CAEV em 16 amostras. Abstract Small ruminant lentiviruses (SRLV) have high genetic variability which results in different viral strains around the world. This create a challenge to design sensible primers for molecular diagnosis in different regions. This work proposes a protocol of duplex nested-PCR for the precise diagnosis of SRLV. The technique was designed and tested with the control strains CAEV Co and MVV 1514. Then, field strains were submitted to the same protocol of duplex nested-PCR. Blood samples of sheep and goats were tested with AGID and nested PCR with specific primers for pol, gag and LTR. Abstract Small ruminant lentiviruses isolated from peripheral blood leukocytes and target organs can be propagated in vitro in fibroblasts derived from goat synovial membrane cells. These cells are obtained from tissues collected from embryos or fetuses and are necessary for the establishment of the fibroblast primary culture. The prevalence of caprine arthritis-encephalitis (CAE) infection in dairy goat herds from Rio Grande do Norte State, Brazil, was estimated, and the predisposing factors to the infection were identified. An epidemiologic survey was held to collect information on educational level of the owner and origin of the animals, and on sanitation and vaccination practices, diseases, morbidity and mortality rates in the herds. The prevalence of CAE virus infection was 11%, and CAE was always observed in some goats in all studied municipalities. |

Este estudo teve como objetivo produzir um antígeno (Ag) a partir de cultura de células de membrana sinovial caprina (MSC) infectadas com o vírus de artrite encefalite caprina (CAEV), pela técnica de microfiltração seriada, substituindo a ultracentrifugação em colchão de sacarose (UCCS) para utilização em ELISA indireto (ELISA-i). Amostras de 188 soros caprinos, que previamente foram testados pelo Western blot (WB) com Ag UCCS, foram submetidas à análise pelo ELISA-i com o novo antígeno produzido, que mostrou concordância de 92% em relação ao antígeno UCCS.
